目的 探讨SH3域结合谷氨酸丰富蛋白样蛋白(SH3BGRL)的表达水平对血管紧张素Ⅱ(AngⅡ)诱导的心肌细胞肥大的影响。方法 通过构建H9C2心肌细胞肥大模型,运用Western blot和RT-PCR技术分别检测SH3BGRL水平,RT-PCR技术检测心肌肥大相关标志基因ANP、BNP和β-MHC的表达情况。结果 过表达SH3BGRL后,心肌肥大相关标志基因的mRNA表达水平升高(P<0.05);而沉默SH3BGRL后,mRNA表达水平显著降低(P<0.05)。结论 SH3BGRL的表达水平与心肌肥大标志基因的表达呈正相关,有望成为治疗心肌肥大的潜在靶点,为深入研究SH3BGRL在心脏中的功能奠定了坚实基础。
Abstract:Objective To investigate the effect of SH3 domain-binding glutamate-rich protein-like protein(SH3BGRL) expression on cardiomyocyte hypertrophy induced by angiotensin Ⅱ(Ang Ⅱ).Methods A hypertrophy model of H9C2 cardiomyocytes was constructed.Western blot and RT-PCR were used to detect the expression level of SH3BGRL,and RT-PCR was used to detect the expression of cardiac hypertrophy-related marker genes ANP,BNP,and β-MHC.Results The mRNA expression levels of cardiac hypertrophy-related marker genes significantly increased after overexpression of SH3BGRL(P<0.05).Conversely, the mRNA expression levels of cardiac hypertrophy-related marker genes significantly decreased after silencing SH3BGRL(P<0.05).Conclusion The expression level of SH3BGRL is positively correlated with the expression of cardiac hypertrophy marker genes, suggesting that it could be a potential target for treating myocardial hypertrophy and laying a solid foundation for further research into the function of SH3BGRL in the heart.
