目的 探讨分泌型卷曲相关蛋白1(SFRP1)对人结直肠癌细胞增殖、侵袭迁移的影响及其机制,为临床治疗提供新的研究方向。方法 使用生物信息学分析SFRP1在结直肠癌中的表达水平及与结直肠癌侵袭和增殖的相关性,研究结直肠癌与SFRP1的关系;将SFRP1抑制物(small interfering RNA)及对照物(si-NC)转染至SW480细胞中;采用CCK-8比色法检测各组细胞增殖能力;通过Transwell实验评估细胞侵袭迁移能力;运用Western blot技术检测各组细胞Wnt/β-catenin信号通路核心组份Wnt4、β-catenin的蛋白表达水平。结果 SFRP1在结直肠癌中表达下调,与对照组SW480细胞相比,SFRP1干扰组细胞增殖,侵袭迁移能力提升,干扰组Wnt4和β-catenin蛋白表达水平增强(P<0.05)。结论 SFRP1表达下调可能通过调控Wnt/β-catenin信号通路,促进结直肠癌的发生发展,为结直肠癌的临床治疗提供了新思路。
Abstract:Objective To investigate the effects of Secreted Frizzled-Related Protein(SFRP1) on the proliferation, invasion, and migration of colorectal cancer cells and its underlying mechanisms, providing a new research direction for clinical treatment.Methods Bioinformatics analysis was used to study the expression level of SFRP1 in colorectal cancer and its correlation with colorectal cancer invasion and proliferation, investigating the relationship between colorectal cancer and SFRP1.SW480 cells were transfected with SFRP1 inhibitor(small interfering RNA) and negtive control(si-NC).CCK-8 colorimetric assay and Transwell assay were performed to evaluate the proliferation, invasion and migration abilities of each group cells.Western blot analysis was used to determine the protein levels of Wnt4 and β-catenin, key components of the Wnt/β-catenin signaling pathway, in each group of cells.Results The expression of SFRP1 protein was down-regulated in colorectal cancer cells(P<0.05).Compared with the control group, the proliferation, invasion and migration ability of si-SFRP1 group enhanced, the expression of Wnt4 and β-catenin protein expressione elevated(P<0.05).Conclusion Downregulation of SFRP1 expression may promote the development and progression of colorectal cancer by regulating the Wnt/β-catenin signaling pathway, providing new insights into the clinical treatment of colorectal cancer.
