摘要:

目的 探究青蒿素(ARS)对大鼠肾缺血/再灌注损伤(RIRI)的作用。方法 将雄性SD大鼠随机分为4组:Sham组、I/R组、I/R+ARS组、ARS组,每组10只。I/R+ARS组和ARS组于手术前5天每天灌胃溶于0.5%羧甲基纤维素溶液(0.5%CMC-Na)的ARS(100mg/kg),而Sham组和I/R组在手术前5天每天灌胃0.5%CMC-Na。I/R组和I/R+ARS组大鼠切除右肾后,用无创动脉夹夹闭左肾肾蒂45min,后松开动脉夹;Sham组和ARS组仅切除右肾,但不阻断肾蒂。术后24h,收集血液及肾组织。Elisa检测血清中Scr、BUN、AST、Kim-1的变化,HE染色观察肾组织损伤情况,Elisa检测肾组织中MDA、SOD、GSH-PX、CAT的变化;IF检测肾组织中NLRP3的表达;Western blot检测NLRP3、Caspase-1、ASC的表达。结果 与Sham组相比,I/R组Scr、BUN、AST、Kim-1显著上升(P<0.05),肾组织出现严重的结构损伤,MDA显著上升(P<0.01),SOD、GSH-PX以及CAT明显下降(P<0.01),NLRP3表达水平上升;而给予ARS预处理后,Scr、BUN、AST、Kim-1显著下降(P<0.05),肾组织结构损伤显著改善,MDA显著下降(P<0.01),SOD、GSH-PX以及CAT明显上升(P<0.05),NLRP3、Caspase-1、ASC表达水平下降(P<0.05)。结论 ARS对大鼠RIRI具有治疗作用,可能与抑制氧化应激,降低NLRP3炎症小体的水平相关。

Abstract:

Objective To investigate the effect of artemisinin(ARS) on renal ischemia/reperfusion injury(RIRI) in rats.Methods Male SD rats were randomly divided into four groups:Sham group,I/R group,I/R+ARS group,and ARS group,with 10 rats in each group.The I/R+ARS group and ARS group were given ARS(100 mg/kg) dissolved in 0.5% carboxymethyl cellulose solution(0.5% CMC-Na) once daily for 5 days before surgery,while the Sham group and I/R group were given 0.5% CMC-Na once daily for 5 days before surgery.In the I/R group and I/R+ARS group,the right kidney was removed,and the left renal pedicle was clamped with a non-invasive arterial clamp for 45 minutes,after which the clamp was released;in the Sham group and ARS group,only the right kidney was removed without clamping the renal pedicle.At 24 hours after surgery,blood and kidney tissues were collected.ELISA was used to detect changes in Scr,BUN,AST,and Kim-1 in serum,HE staining to observe kidney tissue damage,and ELISA to detect changes in MDA,SOD,GSH-PX,and CAT in kidney tissue.IF was used to detect NLRP3 expression in kidney tissue,and Western blot to detect the expression of NLRP3,Caspase-1,and ASC.Results Compared with the Sham group,the I/R group showed significantly increased Scr,BUN,AST,and Kim-1(P<0.01,P<0.01,P<0.01,P<0.05),severe structural damage in kidney tissue,significantly increased MDA(P<0.01),and significantly decreased SOD,GSH-PX,and CAT(P<0.01),with increased NLRP3 expression level.After ARS pretreatment,Scr,BUN,AST,and Kim-1 significantly decreased(P<0.01,P<0.01,P<0.05,P<0.05),structural damage in kidney tissue was significantly improved,MDA significantly decreased(P<0.01),and SOD,GSH-PX,and CAT significantly increased(P<0.01,P<0.01,P<0.05),with decreased expression levels of NLRP3,Caspase-1,and ASC(P<0.05).Conclusion ARS has a therapeutic effect on RIRI in rats,which may be related to inhibiting oxidative stress and reducing the level of NLRP3 inflammasomes.
